Since I am starting from ground zero this may be a funny question for you: Does it matter what screen size you use when you develop the website? I am doing a 1024X726 but I noticed the tutorial used 800 x 600.

Yes, accounting for screen size is very important. When you design for the web, you are designing for a lowest common screen size that you want to support. For years that standard was 800*600, but it has pretty safely moved to 1024*768 these days.

However, keep in mind that if you design for that screen size, the browser takes up some of that space. When I was designing for a 800*600 page, I used to actually do my layouts for a 740*480 space. 960*640 is what I use to design for 1024*768 now.
